Title: Molecular Pattern Formation, Networks, and Graph Theory Can Cause You to Lose Your Mind

Abstract: Catastrophic neurological diseases such as Alzheimer’s and Parkinsons’s occur in the presence of multi-protein aggregates with a specific pattern of hydrogen bonds. The same proteins originate in molecular configurations that are innocuous. These proteins undergo a major structural conversion while converting to the toxic amyloid aggregates. During the structural conversion process, the global H-bond network undergoes a transformation. We propose that the transformation of the H-bond network is the dominant factor in controlling the dynamics of the structural conversion and aggregation. Therefore, determining the details of the transformation of the H-bond network will provide insight on the underlying molecular mechanism of amyloid fibril formation.
